Good idea, lots of possibilities
I like the matching letter variant. It makes movement on the grid and movement rate in a turn a more important aspect.
Each location on the grid could have it's number visible, and a chit is put facedown with the letter of the location it points to. When a clue is revealed at a location, the chit can be turned and players know where to go next. If players need to follow the trail, players can leave a cube or marker in their color to show they visited a location, and can go to the next one.
If during the game a shorter trail to a location becomes available (if some letters appear on different chits), then it makes sense that players take the shorter trail. It might be a good catch up mechanism.
